Side effects, contraindications and regulatory status

Reported side effects

  • FDA page (checked 2026-10-20), kisspeptin-10 in category 2 (503A): compounded versions may pose risk for immunogenicity for certain routes; FDA states it has little or no safety data for the proposed routes.[1] Regulator or expert-panel statement
  • Trial (Hum Reprod 2015, intravenous kisspeptin-10 and -54 vs GnRH in healthy men, five per dosing group): the authors list small sample size as a limitation; the abstract does not report adverse events.[6] Human trial

Regulatory status

FDA 503A: kisspeptin-10 in category 2 (added 2023-09-29), immunogenicity risk for certain routes, little or no safety data for proposed routes. Checked 2026-10-20.[1] Source

What the research says

Summaries of the cited studies, each with its evidence grade.

  • Randomized double-blind parallel trial (J Clin Endocrinol Metab 2009)[2]

    Human trial human

Randomized double-blind parallel trial (J Clin Endocrinol Metab 2009): kisspeptin-54 6.4 nmol/kg by subcutaneous injection twice daily for 2 weeks vs saline in women with hypothalamic amenorrhea (n=5 per group); responses fell by day 14.

    Human clinical trial

  • Single-blind crossover trial (J Clin Endocrinol Metab 2013)[3]

    Human trial human

Single-blind crossover trial (J Clin Endocrinol Metab 2013): kisspeptin-54 6.4 nmol/kg subcutaneously twice daily during menstrual days 7 to 14 vs saline in healthy women (n=5 per arm); menstrual cyclicity persisted in all.

    Human clinical trial

  • pubmed.ncbi.nlm.nih.gov[4]

    Human trial human

Single subcutaneous bolus of kisspeptin-54 0.15, 0.30 or 0.60 nmol/kg or saline in six healthy women in the follicular phase (Clin Endocrinol 2013); LH pulses increased after 0.30 and 0.60 nmol/kg.

    Human clinical trial

  • Kisspeptin-10 (J Clin Endocrinol Metab 2011), n=4 to 5 per group[5]

    Human trial human

Kisspeptin-10 (J Clin Endocrinol Metab 2011), n=4 to 5 per group: intravenous bolus in men and women; subcutaneous bolus and intravenous infusion in women; maximal doses tested in follicular-phase women were 10 nmol/kg IV bolus, 32 nmol/kg SC bolus and 720 pmol/kg/min IV infusion with no gonadotropin change.

    Human clinical trial

  • Single-blind placebo-controlled study (Hum Reprod 2015)[6]

    Human trial human

Single-blind placebo-controlled study (Hum Reprod 2015): kisspeptin-10, kisspeptin-54 and GnRH each infused intravenously for 3 h at 0.1, 0.3 and 1.0 nmol/kg/h in healthy men, five per dosing group, on separate study days at least a week apart.

    Human clinical trial

  • Kisspeptin-10 (J Clin Endocrinol Metab 2011) in healthy men[7]

    Human trial human

    Kisspeptin-10 (J Clin Endocrinol Metab 2011) in healthy men: intravenous bolus 0.01 to 3.0 mcg/kg (maximal LH response at 1 mcg/kg, n=6) and infusions of 1.5 and 4 mcg/kg/h for up to 22.5 h (n=4 each).

    Human clinical trial

  • Eur J Endocrinol 2026 (doi 10.1093/ejendo/lvag134), randomized, single-blinded, placebo-controlled[8]

    Human trial human

Eur J Endocrinol 2026 (doi 10.1093/ejendo/lvag134), randomized, single-blinded, placebo-controlled: acute 8 h subcutaneous kisspeptin-10 infusions of 1.25-10.0 nmol/kg/h raised LH, FSH and testosterone; a continuous 180 nmol/h infusion for 5 days kept testosterone raised but gonadotropins returned to vehicle levels; daily 8 h infusions of 150 nmol/h for 12 days sustained the gonadotropin rise
