Side effects, contraindications and regulatory status

Reported side effects

  • Summary of six placebo-controlled trials (about 900 adults; intravenous and oral routes only; sponsor-affiliated authors): the authors report no drug-related withdrawals or serious adverse events and no anti-AOD9604 antibodies in the subset tested.[2] Human trial
  • In a 7-day oral multiple-dose study (9, 27 or 54 mg daily; 36 obese males) the 54 mg group had more headaches, diarrhea and flatulence than the 9 mg, 27 mg and placebo groups; the authors link the highest dose to more GI-related events.[2] Human trial
  • 24-week oral tablet trial (0.25, 0.5 or 1 mg daily; 502 randomized): 78.7% had at least one adverse event (83.2% placebo, 75.6% at 0.25 mg); 18 patients (3.6%) had a serious event, spread across groups (placebo 8, 0.25 mg 3, 0.5 mg 1, 1 mg 6).[2] Human trial
  • 12-week oral trial (1 to 30 mg daily; 300 obese adults): 5 serious events, all neoplasms (20 mg group: basal cell carcinoma, lipoma, squamous cell carcinoma; 10 mg: malignant melanoma; 5 mg: breast cancer); the investigator judged none related to study medication.[2] Human trial
  • FDA safety-risk page (current 2026-04-22), nominated-but-withdrawn table: FDA has identified no, or only limited, safety-related information, and also serious adverse events that may be associated with AOD-9604, causality not clear.[1] Regulator or expert-panel statement
